كندا لايف – في سابقة هي الأولى من نوعها حقن العلماء فيروساً «قاتلاً للسرطان» في جسم مريض، على أمل أن يكشف الاختبار عن وسيلة جديدة فعالة لمكافحة أورام السرطان.
والعقار المسمى CF33-hNIS، والمعروف أيضاً باسم «فاكسينيا» هو ما يسمى بـ«فيروس محلل الورم»، وهو فيروس معدل وراثياً، صمم لإصابة الخلايا السرطانية وقتلها بشكل انتقائي وتجنب الخلايا السليمة، وذلك حسبما نشر موقع «ساينس إليرت» العلمي المتخصص.
وأظهرت الاختبارات التي أجريت على الحيوانات أنه قادر على تقليل حجم أورام سرطان القولون والرئة والثدي والمبيض والبنكرياس، في حين أن العلاجات المناعية الأخرى كانت فعالة في بعض أنواع السرطان
ويمكن للفيروس تنشيط جهاز المناعة لدى المريض وزيادة مستوى بروتين يسمى PD-L1 في الأورام، ما يجعل العلاج المناعي أكثر فعالية ضد السرطان.
والفيروس الموجود في الطبيعة عُدّل وراثياً على وجه التحديد لمكافحة السرطان من قبل شركة إيموجين للتكنولوجيا الحيوية المتخصصة في العلاجات الجديدة التي تنشط جهاز المناعة ضد السرطان.
وقال دانينغ لي م. د. الباحث الرئيسي والأستاذ المساعد في قسم الأورام الطبية وبحوث المداواة في لوس أنجلوس: «أظهر بحثنا أن فيروسات الأورام يمكن أن تحفز جهاز المناعة على الاستجابة للسرطان وقتله، وكذلك تحفز الجهاز المناعي ليكون أكثر استجابة للعلاجات المناعية الأخرى، بما في ذلك مثبطات نقاط التفتيش»
وأضاف: «حان الوقت الآن لتعزيز قوة العلاج المناعي، ونعتقد أن CF33-hNIS لديه القدرة على تحسين النتائج لمرضانا في معركتهم مع السرطان»
ومن المتوقع أن يشمل الاختبار 100 مريض، في مرحلة متقدمة جداً من تفشي السرطان، والذين تلقوا علاجين تقليديين على الأقل.
وفي هذه التجربة، يعمل الفيروس عبر دخول الخلايا وتكاثره داخلها ما يؤدي إلى انفجار الخلية المصابة وإطلاق آلاف الجزيئات الفيروسية الجديدة التي تحفز جهاز المناعة على مهاجمة الخلايا السرطانية القريبة. ومن المتوقع أن يستمر الاختبار لمدة 24 شهراً تقريباً.
وسيبدأ المرضى بتلقي جرعة منخفضة من «فاكسينيا»، إما عن طريق الحقن مباشرة في الأورام أو عن طريق الوريد.
وسيقيم الباحثون أولاً كيف تتقبل أجسام المرضى هذا الدواء، وسيسجلون الأعراض الجانبية، ومدى تفاعل المشاركين مع زيادة الجرعة.
